How to Read a Peptide COA

A field-by-field guide to sample identity, HPLC purity, measured quantity, chromatograms, lot traceability, and the limits of a certificate—with a publicly verifiable worked example.

Start with the sample—not the percentage.

Before reading a purity result, confirm that the report identifies the same product, strength, batch or lot, and physical sample being evaluated. Then read each result beside its method and specification. A COA can only support the attributes actually tested for the identified sample.

01 · Traceability

Match the package to the analytical record.

A useful record begins with identifiers: product name, stated amount or concentration, batch or lot, report number, testing laboratory, sample-received date, analysis date, and report date. The package and purchasing record should connect to the same lot.

If those identifiers do not align, the document may still be a historical or representative record, but it is not evidence for the physical unit in hand. A polished PDF without sample linkage is not a substitute for traceability.

02 · Method

Ask what the instrument was designed to measure.

High-performance liquid chromatography separates components under defined column, mobile-phase, gradient, temperature, and detection conditions. Reversed-phase HPLC is widely used for peptide separation and characterization. A detector records signals as components leave the column, producing a chromatogram.

Liquid chromatography coupled with mass spectrometry adds mass-selective information. These methods are complementary, not interchangeable. 03 · Separate the claims

Identity, quantity, and purity answer different questions.

Identity

Does the analyzed signal support the intended compound identity under the stated method?

Quantity

How much analyte did the laboratory measure relative to the stated amount or specification?

Chromatographic purity

How dominant was the main detected component relative to other detected peaks under those conditions?

Metals screening

Were named or grouped metals evaluated against a stated reporting limit or specification?

Sterility or endotoxin

Was a separate biological-contamination test actually performed and reported?

Fitness for purpose

Does the laboratory's own qualification show that the material suits its specific experiment?

04 · HPLC purity

What the percentage says—and what it leaves open.

An HPLC purity percentage is method-specific. It summarizes the relative chromatographic signal attributed to the principal component under the stated separation and detector settings. It should be interpreted with the chromatogram, method, integration, and specification.

It is not automatically a measurement of total peptide mass, identity, water, counterions, residual solvents, metals, microbial contamination, endotoxin, or sterility. 05 · Chromatogram

Read the graph behind the result.

The horizontal axis is retention time; the vertical axis is detector response. Peaks represent detected components that reached the detector at different times. The main peak should be considered together with smaller peaks, baseline behavior, integration boundaries, and the reported calculation.

A clean-looking trace does not make every untested attribute true. Method selectivity matters: research on pharmaceutical peptide peak purity shows that column and mobile-phase choices can materially affect separation of closely related peptide species.

06 · Final review

The six-question COA check.

  1. Does the product and batch match the physical package?
  2. Is the testing laboratory named and the report independently verifiable?
  3. Does each result identify a method, unit, specification, and conclusion?
  4. Are identity, measured quantity, and purity treated as separate attributes?
  5. Does the chromatogram or underlying analytical output accompany the summary?
  6. Are untested attributes left unclaimed?

Peptide COA FAQ

Read every field in context.

What should a peptide COA include?

A useful peptide COA identifies the sample or lot, product identity, test method, result, specification, analysis date, testing laboratory, and a way to verify or trace the record. Exact fields vary by method and laboratory.

What does HPLC purity mean on a peptide COA?

HPLC purity is a method-specific chromatographic result based on the separated signals detected under stated conditions. It does not by itself establish identity, total peptide content, sterility, endotoxin status, safety, or suitability for an experiment.

Is peptide identity the same as purity?

No. Identity asks whether the analyzed material matches the intended compound. Purity describes the relative contribution of the main detected component under the stated analytical method. They are separate questions.

Why should the vial lot match the COA?

Matching identifiers connect the physical package to the tested sample and analytical record. A COA from a different sample or historical lot cannot establish the results of the received material.

Does a high HPLC purity result prove a peptide is sterile?
